In vivo studies of PPAR-chromatin interactions: chromatin immunoprecipitation for single-locus and genomewide analyses

Abstract

The immunoprecipitation of protein complexes with associated DNA fragments, termed chromatin immunoprecipitation, allows for the analysis of the binding of endogenous factors. With the advent and widespread use of next generation high-throughput sequencing technologies, factor binding can be assessed in an unbiased and genomewide approach. The protocol outlined here yields sufficient material to subsequently carry out such analyses and discusses key steps in detail.
